Elle peut prendre de nombreuses formes et est difficile \u00e0 diagnostiquer. Si les chercheurs du LCSB souhaitent mieux comprendre les m\u00e9canismes biologiques en jeu, ils sont \u00e9galement constamment \u00e0 la recherche de nouveaux biomarqueurs : des indicateurs susceptibles d\u2019aider \u00e0 identifier les diff\u00e9rents sous-types de la maladie et de permettre un diagnostic pr\u00e9coce. Dans une \u00e9tude r\u00e9cente, des scientifiques du LCSB et des collaborateurs de l\u2019Universit\u00e9 de T\u00fcbingen se sont pench\u00e9s sur le petit fragment d\u2019ADN pr\u00e9sent dans les mitochondries, des composants essentiels des cellules humaines. Les alt\u00e9rations qui s\u2019accumulent dans cet ADN mitochondrial au cours de la vie peuvent \u00eatre d\u00e9tect\u00e9es dans le sang et pourraient constituer une fen\u00eatre sur ce qui se passe dans le cerveau, fournissant ainsi un nouvel outil peu invasif pour faciliter le diagnostic et la m\u00e9decine de pr\u00e9cision.<\/p>\n\n\n\n<h3 class=\"has-text-align-left wp-block-unilux-blocks-heading\"        id=\"les-mitochondries-des-acteurs-cles-de-la-maladie-de-parkinson\"\n    >\nLes mitochondries, des acteurs cl\u00e9s de la maladie de Parkinson<\/h3>\n\n\n\n<p>Chaque cellule humaine contient des mitochondries, des composants sp\u00e9cialis\u00e9s essentiels \u00e0 la survie de nos cellules. Outre leur r\u00f4le dans l\u2019approvisionnement en \u00e9nergie, les mitochondries sont des acteurs cl\u00e9s de notre m\u00e9tabolisme qui r\u00e9gulent aussi l\u2019inflammation. Pour assurer ces fonctions vitales, chaque mitochondrie poss\u00e8de un petit g\u00e9nome circulaire. Ce fragment d\u2019ADN peut accumuler des alt\u00e9rations au cours de la vie, des modifications susceptibles d\u2019avoir un impact consid\u00e9rable sur le fonctionnement de l\u2019organisme et sur le risque de d\u00e9velopper certaines maladies. Ces alt\u00e9rations sont par exemple li\u00e9es \u00e0 certains troubles neurod\u00e9g\u00e9n\u00e9ratifs, en particulier la maladie de Parkinson, et peuvent refl\u00e9ter des changements pr\u00e9coces associ\u00e9s \u00e0 ces maladies.<\/p>\n\n\n\n<p>C\u2019est l\u00e0 qu\u2019interviennent les travaux de recherche men\u00e9s par l\u2019\u00e9quipe <a href=\"https:\/\/www.uni.lu\/lcsb-en\/research-groups\/molecular-functional-neurobiology\/\" target=\"_blank\" rel=\"noreferrer noopener\">Molecular &amp; Functional Neurobiology<\/a> au <a href=\"https:\/\/www.uni.lu\/lcsb-fr\/\" target=\"_blank\" rel=\"noreferrer noopener\">LCSB<\/a> et par ses collaborateurs du <a href=\"https:\/\/www.hih-tuebingen.de\/en\/research\/neurodegenerative-diseases\/research-groups\/fitzgerald-lab\" target=\"_blank\" rel=\"noreferrer noopener\">Fitzgerald Lab<\/a> \u00e0 l\u2019<a href=\"https:\/\/www.hih-tuebingen.de\/en\/\" target=\"_blank\" rel=\"noreferrer noopener\">Hertie Institute for Clinical Brain Research<\/a>. Leur id\u00e9e : d\u00e9terminer si les alt\u00e9rations de l\u2019ADN mitochondrial peuvent \u00eatre d\u00e9tect\u00e9es dans le sang et ainsi constituer un biomarqueur fiable et facilement accessible pour la maladie de Parkinson. Dans leur \u00e9tude la plus r\u00e9cente, <a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/41992946\/\" target=\"_blank\" rel=\"noreferrer noopener\">publi\u00e9e dans la revue scientifique <em>Brain<\/em><\/a>, ils comparent des \u00e9chantillons sanguins provenant de patients atteints de diff\u00e9rentes formes de la maladie de Parkinson, de personnes \u00e0 risque et de volontaires en bonne sant\u00e9.<\/p>\n\n\n\n<h3 class=\"has-text-align-left wp-block-unilux-blocks-heading\"        id=\"des-alterations-de-ladn-pour-identifier-des-formes-specifiques-de-la-maladie-de-parkinson\"\n    >\nDes alt\u00e9rations de l\u2019ADN pour identifier des formes sp\u00e9cifiques de la maladie de Parkinson <\/h3>\n\n\n\n<p>Leurs premiers r\u00e9sultats montrent que, si les alt\u00e9rations de l\u2019ADN mitochondrial sont significativement plus \u00e9lev\u00e9es chez l\u2019ensemble des personnes atteintes de la maladie de Parkinson, la diff\u00e9rence est particuli\u00e8rement frappante pour deux groupes : les patients porteurs de mutations sur deux g\u00e8nes appel\u00e9s <em>PINK1<\/em> et <em>PRKN<\/em>, et d\u2019autre part les patients sans cause g\u00e9n\u00e9tique connue qui d\u00e9veloppent la maladie tr\u00e8s t\u00f4t. \u00ab Ces r\u00e9sultats sont int\u00e9ressants pour deux raisons \u00bb, explique <a href=\"https:\/\/www.uni.lu\/lcsb-en\/people\/anne-grunewald\/\" target=\"_blank\" rel=\"noreferrer noopener\">la professeure Anne Gr\u00fcnewald<\/a>, responsable de l\u2019\u00e9quipe Molecular &amp; Functional Neurobiology. \u00ab Premi\u00e8rement, ils viennent \u00e9tayer l\u2019hypoth\u00e8se d\u2019une implication des mitochondries dans ces deux formes de la maladie de Parkinson, ce qui va orienter les futures \u00e9tudes sur les m\u00e9canismes impliqu\u00e9s dans ces formes-l\u00e0. Ensuite, l\u2019existence de ce nouveau biomarqueur va faciliter la stratification des patients, permettant de les r\u00e9partir en diff\u00e9rents groupes et de mieux adapter les traitements \u00e0 leurs besoins. \u00bb<\/p>\n\n\n\n<h3 class=\"has-text-align-left wp-block-unilux-blocks-heading\"        id=\"un-biomarqueur-precoce-pour-les-personnes-a-haut-risque\"\n    >\nUn biomarqueur pr\u00e9coce pour les personnes \u00e0 haut risque<\/h3>\n\n\n\n<p>De plus, les chercheurs ont observ\u00e9 que les personnes identifi\u00e9es comme \u00e0 risque pour la maladie de Parkinson et celles qui l\u2019ont par la suite d\u00e9velopp\u00e9 pr\u00e9sentaient d\u00e9j\u00e0 des alt\u00e9rations de l\u2019ADN mitochondrial, d\u00e9tectables dans le sang, avant le diagnostic clinique. Ces r\u00e9sultats soulignent que ces alt\u00e9rations sont pr\u00e9sentes d\u00e8s les premiers stades de la maladie de Parkinson et qu\u2019elles refl\u00e8tent sans doute des modifications mol\u00e9culaires pr\u00e9coces associ\u00e9es \u00e0 la maladie. Une approche innovante consisterait donc \u00e0 utiliser un ensemble de marqueurs compl\u00e9mentaires pour identifier la maladie avant l\u2019apparition des premiers sympt\u00f4mes. Pour tester cette hypoth\u00e8se, l\u2019\u00e9quipe a int\u00e9gr\u00e9 diff\u00e9rentes informations portant sur les dommages de l\u2019ADN mitochondrial, sur l\u2019agr\u00e9gation de la prot\u00e9ine alpha-synucl\u00e9ine et sur les scores de risque g\u00e9n\u00e9tiques afin de d\u00e9terminer si cela am\u00e9liorait la capacit\u00e9 \u00e0 distinguer les patients des personnes t\u00e9moins en bonne sant\u00e9.<br> <br>Leurs analyses indiquent que l\u2019analyse des l\u00e9sions de l\u2019ADN mitochondrial fournissent des informations biologiques compl\u00e9mentaires, et qu\u2019une combinaison de plusieurs biomarqueurs offre une repr\u00e9sentation plus compl\u00e8te de l\u2019h\u00e9t\u00e9rog\u00e9n\u00e9it\u00e9 de la maladie de Parkinson.<\/p>\n\n\n\n<ul class=\"wp-block-unilux-blocks-custom-buttons btn-list\"><\/ul>\n<\/div>\n<\/div>\n<\/div>\n<\/section>\n\n\n<section class=\"py-0 wp-block-unilux-blocks-free-section section\"><div class=\"container xl:max-w-screen-xl\">\n<p>\u00ab Ce que nous constatons, c\u2019est que des biomarqueurs distincts nous renseignent sur diff\u00e9rentes dimensions de la maladie \u00bb, explique <a href=\"https:\/\/www.linkedin.com\/in\/julia-fitzgerald-73a17370\/?locale=en\" target=\"_blank\" rel=\"noreferrer noopener\">Dr Julia Fitzgerald<\/a>, co-auteure de l\u2019\u00e9tude. \u00ab Par exemple, certaines mesures mettent en \u00e9vidence l\u2019impact des mutations g\u00e9n\u00e9tiques tandis que d\u2019autres soulignent le r\u00f4le des facteurs environnementaux. Certains marqueurs indiquent ce qui se passe dans le cerveau, d\u2019autres r\u00e9v\u00e8lent l\u2019impact de la neurod\u00e9g\u00e9n\u00e9rescence \u00e0 la p\u00e9riph\u00e9rie de l\u2019organisme. Pris ensemble, ils refl\u00e8tent bien mieux la r\u00e9alit\u00e9 de la maladie. \u00bb<\/p>\n\n\n\n<p>En rassemblant les informations fournies par ces diff\u00e9rents biomarqueurs, les chercheurs pourront non seulement mieux \u00e9tudier les m\u00e9canismes biologiques impliqu\u00e9s dans la maladie, mais ils pourront \u00e9galement d\u00e9velopper une approche multimodale pour faciliter le diagnostic et la m\u00e9decine de pr\u00e9cision.<\/p>\n\n\n\n<h3 class=\"has-text-align-left wp-block-unilux-blocks-heading\"        id=\"une-decennie-de-recherche-collaborative-porte-ses-fruits\"\n    >\nUne d\u00e9cennie de recherche collaborative porte ses fruits<\/h3>\n\n\n\n<p>Ensemble, ces r\u00e9sultats positionnent les alt\u00e9rations de l\u2019ADN mitochondrial comme un nouvel atout&nbsp;: un biomarqueur d\u00e9tectable d\u00e8s les stades pr\u00e9coces de la maladie, dans des \u00e9chantillons sanguins faciles \u00e0 pr\u00e9lever, sp\u00e9cifique \u00e0 certaines formes de la maladie et pouvant \u00eatre combin\u00e9 avec des biomarqueurs g\u00e9n\u00e9tiques et prot\u00e9iques pour une meilleure stratification des patients.<\/p>\n\n\n\n<p>Cette \u00e9tude met \u00e9galement en \u00e9vidence l\u2019importance de la recherche au long cours et des collaborations scientifiques, comme le souligne la professeur Gr\u00fcnewald : \u00ab Ce qui est vraiment passionnant, c\u2019est que nous \u00e9tablissons d\u00e9sormais des liens entre diff\u00e9rents projets men\u00e9s au cours de la derni\u00e8re d\u00e9cennie par le LCSB et diff\u00e9rents collaborateurs, de nos travaux sur les <a href=\"https:\/\/www.uni.lu\/lcsb-fr\/news\/scores-de-risque-genetiques-et-medecine-de-precision-pour-parkinson\/\" target=\"_blank\" rel=\"noreferrer noopener\">scores de risque polyg\u00e9niques<\/a> \u00e0 la <a href=\"https:\/\/www.uni.lu\/fr\/news\/decouverte-inedite-pour-le-diagnostic-des-maladies-neurodegeneratives\/\" target=\"_blank\" rel=\"noreferrer noopener\">m\u00e9thode permettant de d\u00e9tecter des fibrilles d\u2019alpha-synucl\u00e9ine dans le sang<\/a> d\u00e9velopp\u00e9 par des chercheurs japonais en collaboration avec des scientifiques luxembourgeois, en passant par notre exploration du r\u00f4le des mitochondries dans la neurod\u00e9g\u00e9n\u00e9rescence.
