Dans une \u00e9tude r\u00e9cente, deux \u00e9quipes du Luxembourg Centre for Systems Biomedicine<\/a> (LCSB) de l’Universit\u00e9 du Luxembourg, ainsi que des partenaires nationaux et internationaux, ont collabor\u00e9 pour explorer le r\u00f4le de l’alimentation et du microbiote intestinal dans la progression de la maladie de Parkinson. Leurs r\u00e9sultats, publi\u00e9s dans la revue scientifique Cell Reports<\/a>, montrent qu\u2019une carence en fibres alimentaires et l’exposition \u00e0 une prot\u00e9ine bact\u00e9rienne sp\u00e9cifique peuvent exacerber la maladie. Ils soulignent l’importance d\u2019un mode de vie adapt\u00e9 pour les personnes vivant avec la maladie de Parkinson.<\/p>\n\n\n\n

La maladie de Parkinson est un trouble neurologique caract\u00e9ris\u00e9 par la perte de certains neurones dans le cerveau et l’agr\u00e9gation d’une prot\u00e9ine humaine appel\u00e9e \u03b1-synucl\u00e9ine. Si plusieurs facteurs de risque pour cette maladie sont assez bien connus, il reste encore beaucoup \u00e0 faire pour d\u00e9terminer ce qui peut moduler son \u00e9volution dans le temps. Dans ce contexte, des chercheurs des \u00e9quipes \u2018Systems Ecology<\/a>\u2019 et \u2018Neuropathology<\/a>\u2019 du LCSB ont alli\u00e9 leurs expertises pour \u00e9tudier les effets du r\u00e9gime alimentaire, de la composition du microbiote intestinal et des prot\u00e9ines s\u00e9cr\u00e9t\u00e9es par ces microorganismes dans un mod\u00e8le murin de la maladie de Parkinson.<\/p>\n\n\n\n

\u00ab Sur la base d’une s\u00e9rie de tests, nous avons d\u00e9termin\u00e9 qu’une lign\u00e9e de souris transg\u00e9niques sp\u00e9cifique, reproduisant les principales caract\u00e9ristiques de la maladie de Parkinson, constituait un mod\u00e8le appropri\u00e9 pour \u00e9tudier l’impact des facteurs environnementaux, tels que le r\u00e9gime alimentaire, sur l’\u00e9volution de cette maladie, \u00bb explique Dr Kristopher Schmit<\/a>, qui a men\u00e9 l’\u00e9tude dans le cadre de son doctorat au sein des deux groupes de recherche. \u00ab Ces souris surexpriment la prot\u00e9ine \u03b1-synucl\u00e9ine, ce qui entra\u00eene l’apparition progressive de sympt\u00f4mes moteurs, comme chez les patients. \u00bb<\/p>\n\n\n\n

Lorsque ces souris transg\u00e9niques ont \u00e9t\u00e9 nourries avec un r\u00e9gime pauvre en fibres, la diversit\u00e9 microbienne au niveau de leur intestin s\u2019est appauvrie. Plus particuli\u00e8rement, certaines bact\u00e9ries ayant des fonctions neuroprotectrices ont d\u00e9clin\u00e9, tandis que d\u2019autres qui se nourrissent de mucus ont prosp\u00e9r\u00e9, entra\u00eenant un amincissement de la couche externe de mucus dans le c\u00f4lon. \u00ab Le mucus agit comme une barri\u00e8re physique contre les pathog\u00e8nes. Son \u00e9rosion, provoqu\u00e9e par les changements au niveau de la flore intestinale, pourrait donc faciliter l’acc\u00e8s de ces agents pathog\u00e8nes aux couches plus profondes de l’intestin, o\u00f9 se situent notamment des cellules nerveuses, \u00bb explique Prof. Paul Wilmes<\/a>, responsable de l\u2019\u00e9quipe Systems Ecology.<\/p>\n\n\n\n

L\u2019int\u00e9grit\u00e9 de la barri\u00e8re intestinale, d\u00e9j\u00e0 alt\u00e9r\u00e9e par une carence en fibres, est davantage perturb\u00e9e si le r\u00e9gime alimentaire d\u00e9s\u00e9quilibr\u00e9 se double d\u2019une exposition \u00e0 la prot\u00e9ine curli. Cette derni\u00e8re est produite par des bact\u00e9ries comme Escherichia coli, souvent pr\u00e9sente chez les personnes atteintes de la maladie de Parkinson, et peut acc\u00e9l\u00e9rer l’agr\u00e9gation de l’\u03b1-synucl\u00e9ine humaine. Ainsi, chez les souris transg\u00e9niques, la pr\u00e9sence de curli a entra\u00een\u00e9 une augmentation des agr\u00e9gats d’\u03b1-synucl\u00e9ine dans les nerfs p\u00e9riph\u00e9riques qui peuplent le c\u00f4lon. \u00ab Nous avons par la suite pu constater qu’en plus d’affecter l’intestin lui-m\u00eame, ce processus s’est propag\u00e9 au cerveau, \u00bb poursuit le professeur Wilmes.<\/p>\n\n\n

\n \n\"Repr\u00e9sentation <\/figure>\n\n\n

En effet, les chercheurs de l\u2019\u00e9quipe Neuropathology ont ensuite explor\u00e9 l’impact de ce double traitement sur les capacit\u00e9s motrices et les neuropathologies des souris transg\u00e9niques. Ils ont d’abord observ\u00e9 que les d\u00e9ficits moteurs \u00e9taient exacerb\u00e9s chez les souris expos\u00e9es \u00e0 la prot\u00e9ine curli et \u00e0 un r\u00e9gime \u00e0 faible teneur en fibres, celles-ci montrant une capacit\u00e9 de coordination r\u00e9duite au fil du temps. De m\u00eame, l’agr\u00e9gation d’\u03b1-synucl\u00e9ine s\u2019est r\u00e9v\u00e9l\u00e9e plus importante dans leur cerveau. \u00ab En \u00e9valuant la perte en neurones, nous avons conclu que la pr\u00e9sence de la prot\u00e9ine curli dans l\u2019intestin combin\u00e9 \u00e0 une carence en fibres \u00e9tait un moteur de la neurod\u00e9g\u00e9n\u00e9rescence dans ce mod\u00e8le de la maladie de Parkinson, \u00bb explique Dr Manuel Buttini<\/a>, chercheur dans le groupe Neuropathology dirig\u00e9 par le professeur Michel Mittelbronn.<\/p>\n\n\n\n

Les auteurs d\u00e9crivent le m\u00e9canisme qui pourrait entrer en jeu : \u00ab Pour expliquer nos r\u00e9sultats, nous proposons la s\u00e9quence suivante : une carence chronique en fibres alimentaires entra\u00eene des changements au sein des populations microbiennes pr\u00e9sentes dans l\u2019intestin. Cela d\u00e9clenche ensuite une \u00e9rosion du mucus et une plus grande perm\u00e9abilit\u00e9 de la paroi intestinale, facilitant l’entr\u00e9e d’agents pathog\u00e8nes. Plus particuli\u00e8rement, l’exposition des neurones ent\u00e9riques \u00e0 des prot\u00e9ines telles que curli amplifie l\u2019agr\u00e9gation de l’\u03b1-synucl\u00e9ine dans le syst\u00e8me nerveux ent\u00e9rique et central, entra\u00eenant une neurod\u00e9g\u00e9n\u00e9rescence accrue et une exacerbation des d\u00e9ficits moteurs. \u00bb<\/p>\n\n\n\n

L’\u00e9tude apporte ainsi un \u00e9clairage important sur la mani\u00e8re dont le r\u00e9gime alimentaire et son action sur le microbiote intestinal peuvent moduler la progression de la maladie de Parkinson par l’interm\u00e9diaire de l’axe intestin-cerveau. Elle a \u00e9galement des implications en mati\u00e8re d\u2019adaptation du mode de vie pour les patients. En effet, des mesures simples, comme un r\u00e9gime alimentaire \u00e9quilibr\u00e9 et une utilisation contr\u00f4l\u00e9e des antibiotiques afin de ne pas favoriser la prolif\u00e9ration de bact\u00e9ries produisant la prot\u00e9ine curli, pourraient contribuer \u00e0 ralentir la progression de la maladie.<\/p>\n\n\n\n
